Rat Eae Model

The rat EAE model is an animal model of experimental autoimmune encephalomyelitis, an induced inflammatory disease of the central nervous system used to study immune-mediated demyelination and multiple sclerosis. Researchers typically immunize rats with myelin-associated antigens, often with an adjuvant, to activate autoreactive T cells that cross the blood-brain barrier and promote inflammation, myelin injury, and neurological deficits. Disease progression can be assessed through clinical scoring, behavioral changes, histopathology, and immune responses. This model supports investigation of neuroinflammatory mechanisms and evaluation of potential therapies, while its species-specific immune and disease features must be considered when interpreting results.

Determining Blood-Brain Barrier Disruption Using Fluorescent Dyes in an EAE Mouse Model

This video demonstrates a procedure for assessing blood-brain barrier disruption in an EAE mouse model. Fluorescent dyes of different sizes are injected retro-orbitally and circulate through the brain's blood vessels. The smaller dye leaks through gaps into the brain tissue, while the larger dye enters only if the disruption is severe, correlating with the extent of barrier damage.

The transplantation of neural stem/progenitor cells (NPCs) holds great promises in regenerative neurology. The systemic delivery of NPCs has turned into effective, low invasive, and therapeutically very efficacious protocol to deliver stem cells in the brain and spinal cord of rodents and nonhuman primates affected by experimental chronic inflammatory damage of the central nervous system.
