Recipient Preparation

Recipient preparation is the set of biological and clinical steps used to ready a host for receiving transplanted cells, tissues, or organs, helping establish conditions for successful engraftment and function. In transplantation biology, preparation may involve conditioning treatments that reduce immune activity, remove or suppress competing host cells, and create a receptive tissue environment before donor material is introduced. Careful recipient preparation can improve donor-cell survival, limit rejection, and support tissue reconstitution, making it important in hematopoietic stem cell transplantation, regenerative medicine, and experimental transplant models. Its design depends on the graft type, recipient condition, and balance between treatment effectiveness and toxicity.

This video demonstrates an assay to detect anti-donor HLA alloantibodies in organ recipients. A peptide array is synthesized to represent potential epitopes for antibody-mediated organ rejection. The array is incubated with the recipient's serum, containing alloantibodies that bind to their target epitopes. Antibody binding is detected using chemiluminescence to identify donor-specific HLA epitopes that incite alloantibody reactivity.
