Staple-scaffold Annealing

Staple-scaffold annealing is a molecular self-assembly method used to fold a long DNA scaffold strand into defined nanostructures with many shorter staple strands. During controlled cooling, complementary base pairing causes each staple to bind selected regions of the scaffold, while neighboring staples stabilize the programmed shape and geometry. In biology, this approach supports the construction of DNA origami objects, nanoscale platforms, and spatially organized assemblies for studying molecular interactions, presenting biomolecules, and designing tools for biosensing or delivery. Annealing conditions influence hybridization efficiency, structural yield, and the accuracy of the final nanostructure.
