Sterile Medium Preparation

Sterile medium preparation is the process of producing nutrient-rich culture media free from viable microbial contaminants, a critical foundation for reliable biological experiments. It typically involves dissolving defined components, adjusting pH and volume, sterilizing the medium by autoclaving or membrane filtration when appropriate, and transferring it under aseptic conditions to preserve sterility. Proper preparation supports the controlled growth of cells, tissues, or microorganisms in research and biotechnology. Consistent composition and contamination control improve experimental reproducibility, enable accurate assessment of growth or treatment responses, and help maintain healthy cultures for applications such as cell biology, microbiology, and tissue engineering.

As obligate anaerobic organisms are unable to grow upon oxygen exposure, the use of anaerobic culturing techniques is indispensable. Here, we demonstrate a simple and effective method to cultivate a mixed culture derived from a biogas plant from media preparation to gas and volatile fatty acid quantification.
