Synthetic Mirnas

Synthetic miRNAs are laboratory-designed short RNA molecules that mimic endogenous microRNAs and regulate gene expression through sequence-specific post-transcriptional silencing. After entering a cell, they are incorporated into the RNA-induced silencing complex, whose Argonaute protein uses the miRNA sequence to recognize complementary messenger RNA, promoting transcript degradation or inhibiting translation. In biology, synthetic miRNAs help researchers reduce expression of selected genes and investigate gene function, signaling pathways, and disease mechanisms. They also support therapeutic research, including approaches for cancer, genetic disorders, and tissue regeneration, although delivery, stability, and unintended off-target effects remain important challenges.

This video describes the extraction of circulating microRNA from plasma samples of colorectal cancer patients using organic extraction followed by glass-fiber column enrichment. miRNAs are stable in biological fluids such as serum and plasma, which renders them ideal circulating biomarkers for cancer diagnosis, prognosis, and treatment decision-making and monitoring. This method will help identify new circulating biomarkers capable of predicting clinical outcomes in patients.
