Binary Vector Plasmid

A binary vector plasmid is a DNA construct used to introduce and express selected genes in plant cells, typically through Agrobacterium-mediated transformation. It carries the gene of interest and selectable markers between right and left T-DNA border sequences, while a separate helper plasmid supplies virulence proteins that process and transfer this bordered DNA into the plant cell. Once transferred, the T-DNA can integrate into the plant genome and direct transgene expression. Binary vectors support plant biotechnology applications including functional genomics, crop improvement, protein production, and the development of genetically engineered plants.

Using a pBIBAC-GW binary vector makes generating transgenic plants with intact single-copy insertions, an easy process. Here, a series of protocols is presented that guide the reader through the process of generating transgenic Arabidopsis plants, and testing the plants for intactness and copy number of the inserts.

Plasmid Purification

2023

Plasmid purification is a technique used to isolate and purify plasmid DNA from genomic DNA, proteins, ribosomes, and the bacterial cell wall. A plasmid is a small, circular, double-stranded DNA that is used as a carrier of specific DNA molecules. When introduced into a host organism via transformation, a plasmid will be replicated, creating numerous copies of the DNA fragment under study. In this video, a step-by-step generalized procedure is described for how to perform plasmid purification.

Binary Fission

Binary fission is the primary mode of asexual reproduction in prokaryotes, such as bacteria. It results in the production of two genetically identical daughter cells. This highly efficient process ensures the rapid propagation of bacterial populations under favorable conditions and involves coordinated cellular and molecular events.DNA Replication and SeparationThe process begins with the replication of the bacterial chromosome. The circular DNA molecule unwinds at a specific origin of...
