Tau Protein

Tau protein is a microtubule-associated protein that helps maintain the structure and organization of neuronal axons, making it important for normal brain function. In healthy neurons, tau binds to and stabilizes microtubules, supporting intracellular transport; abnormal phosphorylation can reduce this binding and promote tau misfolding and aggregation into neurofibrillary tangles. These aggregates are characteristic of Alzheimer’s disease and other neurodegenerative tauopathies, where they are associated with neuronal dysfunction and loss. Studying tau protein supports research into disease mechanisms, fluid and imaging biomarkers, and potential therapies aimed at preventing abnormal modification, aggregation, or spread.

In Vitro Aggregation Assays Using Hyperphosphorylated Tau Protein

Unmodified and hyperphosphorylated tau proteins were used in two in vitro aggregation assays to reveal the hyperphosphorylation-dependent fast aggregation kinetics. These assays pave the way for future screens for compounds that can modulate the propensity of hyperphosphorylated tau to form fibrils that underlie the progression of Alzheimer’s disease.
