Triton X-100 Treatment

Triton X-100 treatment is a laboratory method that uses a nonionic detergent to disrupt biological membranes while helping preserve many proteins and cellular structures. Its hydrophobic portion interacts with membrane lipids, whereas its hydrophilic portion maintains detergent-lipid complexes in solution, enabling controlled membrane permeabilization or cell lysis depending on concentration, exposure time, and sample type. In biology, researchers use Triton X-100 to prepare cells for immunofluorescence, improve antibody access to intracellular targets, extract membrane-associated proteins, and separate soluble from insoluble cellular components. Careful optimization is essential because excessive detergent exposure can damage morphology or remove target molecules.

This video demonstrates the immunohistochemical staining for S-100 protein in rectal suction biopsy sections. Antigen-retrieved sections are blocked and treated with primary, followed by secondary antibodies against S-100, a marker of Hirschsprung's disease. Chromogenic substrate is added to stain the targeted cells. Samples are dehydrated and mounted before visualizing under the microscope.
