Tryptophan-containing Peptides

Tryptophan-containing peptides are short chains of amino acids that include tryptophan, an aromatic residue whose indole ring contributes distinctive structural, optical, and chemical properties. The indole group can participate in hydrophobic interactions, hydrogen bonding, and cation–π interactions, while its fluorescence provides a sensitive probe of peptide conformation and the local environment. In biology, these peptides help researchers investigate protein folding, membrane association, ligand binding, and molecular recognition. Their sequence-dependent behavior also supports applications in peptide design, biosensor development, fluorescence-based assays, and studies of how amino acid composition influences biological function.

Measuring Peptide Translocation into Large Unilamellar Vesicles

This protocol details a method for the quantitative measure of peptide translocation into large unilamellar lipid vesicles. This method also provides information about the rate of membrane translocation and can be used to identify peptides that efficiently and spontaneously cross lipid bilayers.

ANS binds to the Ca2+-ATPase recombinant N-domain. Fluorescence spectra display a FRET-like pattern upon excitation at a wavelength of 295 nm. NBS-mediated chemical modification of Trp quenches the fluorescence of the N-domain, which leads to the absence of energy transfer (FRET) between the Trp residue and ANS.
