Valvular Cushion Isolation

Valvular cushion isolation is a developmental biology technique used to obtain the embryonic heart tissues that give rise to cardiac valves, enabling researchers to study how these structures form and mature. The method relies on precise microscopic dissection of valvular cushions from developing hearts, followed by tissue separation, culture, or molecular analysis while preserving their cellular and extracellular components. Isolated cushions provide a controlled system for examining endocardial cushion formation, epithelial-to-mesenchymal transition, cell differentiation, and matrix remodeling. This approach supports research into congenital heart defects, valve development, and the cellular mechanisms that shape normal and abnormal cardiac anatomy.

Classically, the endocardium of the mouse embryonic valve primordium has been analyzed using transversal, coronal, or sagittal sections. Our novel approach for en face, two-dimensional imaging of the endocardium at valvulogenic regions allows planar polarity and cell rearrangement analysis of the endocardium during valve development.
