Vesicle Priming

Vesicle priming is the cellular process that prepares secretory vesicles for rapid, regulated fusion with the plasma membrane, enabling efficient release of neurotransmitters, hormones, or other cargo. In synapses, priming occurs after vesicles are recruited and docked at active zones, where interactions among SNARE proteins and associated factors organize a fusion-ready state. When calcium enters through voltage-gated channels, it binds a vesicle calcium sensor and triggers rapid membrane fusion and exocytosis. Studying vesicle priming clarifies how synaptic strength, secretion timing, and short-term plasticity are controlled, while linking defects in release machinery to neurological and secretory disorders.

This technique provides a method to harvest, normalize and quantify intracellular growth of bacterial pathogens that are pre-cultivated in natural protozoan host cells prior to infections of mammalian cells. This method can be modified to accommodate a wide variety of host cells for the priming stage as well as target cell types.
