Volume Measurement

Volume measurement is the determination of the three-dimensional space occupied by a substance, solution, specimen, or biological structure, providing a basis for accurate quantitative work in biology. It relies on calibrated instruments such as graduated cylinders, pipettes, and volumetric flasks, with readings taken at the liquid meniscus; irregular objects can be measured by fluid displacement, while regular shapes may be calculated from their dimensions. Accurate volume measurement supports solution preparation, dilution series, microscopy-based size analysis, and comparisons of tissue or cell samples, helping researchers control experimental conditions and interpret biological changes reproducibly.

Here we present a protocol to simply and reliably measure the lung pressure-volume curve in mice, showing that it is sufficiently sensitive to detect phenotypic parenchymal changes in two common lung pathologies, pulmonary fibrosis and emphysema. This metric provides a means to quantify the lung’s structural changes with developing pathology.
