Crypt Detachment Pipetting

Crypt detachment pipetting is a mechanical technique used to release intestinal epithelial crypts from surrounding tissue for downstream analysis or culture. It matters because isolating intact crypt units can preserve multicellular organization that supports studies of epithelial biology and cancer. During the procedure, researchers repeatedly aspirate and dispense a tissue suspension through a pipette, applying controlled shear forces that separate crypts while limiting disruption of their structure. In cancer research, this approach can help obtain normal or tumor-associated crypt populations for organoid establishment, molecular profiling, and testing how mutations or treatments affect growth and tissue organization.

Here, we present a protocol for photoelectron imaging of anionic species. Anions generated in vacuo and separated by mass spectrometry are probed using velocity mapped photoelectron imaging, providing details of anion and neutral energy levels, anion and neutral structure and the nature of the anion electronic state.

In this protocol, we describe how to generate murine primary epithelial colon monolayers directly from intestinal crypts. We provide experimental approaches to generate confluent monolayers on permeable filters, confluent monolayers for scratch wound healing and biochemical studies, and sparse and confluent monolayers for immunofluorescence analysis.
