Embryo Recovery

Embryo recovery is the collection of preimplantation embryos from a reproductive tract or culture system, a foundational procedure for studying development and generating experimental models. In laboratory animals, researchers typically flush the oviduct or uterus with sterile, embryo-compatible medium, identify embryos under a stereomicroscope, and assess their developmental stage and morphology before culture, cryopreservation, genotyping, or transfer. In cancer research, recovered embryos can support the production of genetically engineered animals carrying oncogenic mutations, enable early assessment of developmental effects, and help preserve valuable disease-model lines. Standardized recovery improves embryo viability and reproducibility, strengthening studies of tumor initiation, inherited cancer susceptibility, and targeted therapies.

2011

Here we demonstrate the newly developed I•Cryo kit for mouse sperm cryopreservation. Two-cell stage embryo development with frozen-thawed sperm was improved consistently in 5 mouse strains with the use of this kit. Over a 1.5 year period, 49 genetically modified mouse lines were archived by sperm cryopreservation with the I•Cryo kit and later successfully recovered by IVF.

2023

This video describes how to perform microinjections that deliver reagents into live Drosophila embryos. Researchers can use this method to introduce exogenous compounds like nucleic acids and proteins during the early stages of embryo development. The featured protocol demonstrates how to set up and perform the procedure for injection of any soluble reagent–here, a fluorescent protein for live imaging experiments.
