Microdroplet Pcr

Microdroplet PCR is a digital polymerase chain reaction technique that partitions a DNA sample into thousands of tiny, independent droplets, enabling highly sensitive and quantitative nucleic acid analysis. Each droplet functions as an individual reaction chamber, and after thermal cycling, fluorescence identifies droplets containing amplified target sequences; statistical analysis of positive and negative droplets estimates the starting copy number through Poisson distribution. In cancer research, microdroplet PCR supports the detection of rare mutations, copy-number changes, pathogens, and circulating tumor DNA within complex samples. Its high sensitivity and tolerance for small concentration differences can improve biomarker analysis, treatment monitoring, and minimal residual disease assessment.

Capillary-based Centrifugal Microfluidic Device for Size-controllable Formation of Monodisperse Microdroplets

Here, we demonstrate a simple production method for size-controllable, monodisperse, water-in-oil (W/O) microdroplets using a capillary-based centrifugal microfluidic device. This method requires only a small sample volume and enables high-yield production. We expect this method will be useful for rapid biochemical and cellular analyses.
