Microrna Sequencing

MicroRNA sequencing is a next-generation sequencing method that identifies and quantifies small, regulatory RNA molecules, providing a detailed view of gene regulation in cells and tissues. The workflow typically converts mature microRNAs into sequencing libraries through adapter ligation, reverse transcription, and amplification, followed by high-throughput sequencing and computational alignment to reference genomes or databases. In cancer research, these profiles can reveal tumor-associated microRNA signatures, support comparisons between normal and malignant tissues, and help investigate pathways involved in proliferation, invasion, metastasis, and treatment response. The resulting data can guide biomarker discovery and improve understanding of post-transcriptional regulation in cancer.

MicroRNA (miRNA) are short, regulatory RNA transcribed from introns—non-coding regions of a gene—or intergenic regions—stretches of DNA present between genes. Herein we propose a strategy to study the effect of a transcription factor of interest on the microRNA transcriptome using publically available data, computational resources and high throughput data from microRNA arrays after transfecting cells with small hairpin (sh)RNA targeting a transcription factor of interest.
