Orthotopic Engraftment

Orthotopic engraftment is the implantation of cancer cells or tumor tissue into the anatomically corresponding organ of a model organism, preserving site-specific features that influence tumor behavior. After implantation, engrafted cells interact with the native stroma, vasculature, extracellular matrix, and local signaling environment, supporting tumor establishment and progression in a context that more closely reflects disease in patients. In cancer research, orthotopic xenograft and patient-derived tumor models help investigators study local invasion, metastasis, tumor biology, and therapeutic response. These models can improve preclinical evaluation of anticancer treatments by capturing tissue-specific effects that may be missed by tumors grown at nonnative sites.

Pre-Conditioning the Airways of Mice with Bleomycin Increases the Efficiency of Orthotopic Lung Cancer Cell Engraftment

We describe a method to significantly enhance orthotopic engraftment of lung cancer cells into the murine lungs by pre-conditioning the airways with injury. This approach may also be applied to study stromal interactions within the lung microenvironment, metastatic dissemination, lung cancer co-morbidities, and to more efficiently generate patient derived xenografts.

An Orthotopic Sciatic Nerve Xenograft for Neurofibromatosis Type 1 Neurofibromas

Neurofibromatosis type 1 (NF1) leads to benign peripheral nerve sheath tumors (neurofibromas) that require a specific nerve microenvironment for growth. An optimized orthotopic xenograft model was developed, using intraneural injection of human Schwann cells into NSG mice. This model recapitulates human neurofibromas and provides a valuable platform to evaluate therapeutic strategies.

Mantle cell lymphoma (MCL) is a difficult to treat B cell disorder and it is equally difficult to establish a xenograft mouse model of primary MCL to study and develop therapeutics. Here, we describe the successful establishment of MCL xenografts in mice to help understand its underlying biology.
