Orthotopic Injection

Orthotopic injection is a cancer-modeling technique that places tumor cells or tissue into the anatomical site where the cancer naturally originates. Unlike ectopic implantation, it preserves relevant local interactions with surrounding stromal cells, extracellular matrix, vasculature, and immune components. By introducing cells into the appropriate organ under controlled conditions, researchers can reproduce aspects of tumor growth, invasion, treatment response, and spontaneous metastasis that depend on the tissue microenvironment. Orthotopic injection supports preclinical studies of cancer biology, drug efficacy, imaging, and surgical strategies, while helping investigators assess how tumors interact with their native host tissue.

2023

The video describes the technique for intracecal injection of cancerous cells to develop an orthotopic mouse model with highly uniform tumors. Therefore, the orthotopic mouse model allows realistic simulation of metastasis, providing a valuable tool to study the phenotype changes during tumor growth and dissemination.
