Organic Extraction Method

Organic extraction is a sample-preparation method that separates compounds from a complex mixture by transferring them into an organic solvent, making it important for chemical and biological analysis. The process typically uses differences in solubility and polarity: a sample is mixed with an immiscible organic phase, target molecules partition between solvent layers, and the phases are separated before the extract is dried or concentrated. In cancer research, organic extraction helps isolate lipids, metabolites, and potential bioactive compounds from cells, tissues, or natural products. These extracts support chromatography, compound characterization, biomarker studies, and evaluation of substances with possible anticancer activity.

Circulating miRNA Extraction: A Method to Isolate miRNA from Plasma Samples Via Organic Extraction and Small RNA Enrichment

2023

This video describes the extraction of circulating microRNA from plasma samples of colorectal cancer patients using organic extraction followed by glass-fiber column enrichment. miRNAs are stable in biological fluids such as serum and plasma, which renders them ideal circulating biomarkers for cancer diagnosis, prognosis, and treatment decision-making and monitoring. This method will help identify new circulating biomarkers capable of predicting clinical outcomes in patients.

Metal ions can be separated from one another by complexation with organic ligands–the chelating agent– to form uncharged chelates. Here, the chelating agent must contain hydrophobic groups and behave as a weak acid, losing a proton to bind with the metal. Since most organic ligands used in this process are insoluble or undergo oxidation in the aqueous phase, the chelating agent is initially added to the organic phase and extracted into the aqueous phase. The metal-ligand complex is formed in...
