Pancreatic Tumor Microenvironment

The pancreatic tumor microenvironment is the network of cells, extracellular matrix, blood vessels, and signaling molecules surrounding and interacting with pancreatic cancer cells. It influences tumor behavior through reciprocal communication, in which cancer cells recruit and reprogram fibroblasts, immune cells, and vascular components while matrix remodeling alters tissue stiffness, nutrient access, and drug penetration. In cancer research, studying this microenvironment helps explain tumor growth, immune suppression, metastasis, and resistance to treatment. Experimental models that incorporate stromal and immune components can support the evaluation of targeted therapies and improve understanding of why pancreatic tumors often respond poorly to conventional treatments.

Pancreatic juice is a precious source of biomarkers for human pancreatic cancer. We describe here a method for intraoperative collection procedure. To overcome the challenge of adopting this procedure in murine models, we suggest an alternative sample, tumor interstitial fluid, and describe here two protocols for its isolation.

Microfluidic Device for Recreating a Tumor Microenvironment in Vitro

We present the procedure for fabrication and operation of a microfluidic device that recreates heterogeneous tumor microenvironments in vitro. The variability in apoptosis within tumor tissue was quantified using fluorescent stains and the effective diffusion coefficient of the chemotherapeutic drug doxorubicin into tumor tissue was evaluated.

PTM Based Tumor Microenvironment Monitoring: A Method for Tracking Changes in ECM Rigidity Using Tumor Spheroids

This protocol describes a technique for monitoring extracellular matrix rigidity changes by using particle-tracking microrheology (PTM). This is done by culturing 3D pancreatic tumor spheroids into ECM with embedded fluorescent probes for video particle-tracking and analysis.
