Radiotherapy Biomarkers

Radiotherapy biomarkers are measurable biological, molecular, or imaging features that indicate how a tumor or normal tissue may respond to radiation, helping guide personalized cancer treatment. They work by reflecting processes such as tumor hypoxia, DNA damage and repair, cell-cycle regulation, or immune activity that influence radiosensitivity and treatment toxicity. In cancer research, these biomarkers support patient selection, response prediction, treatment monitoring, and risk assessment for side effects. Their development may improve radiation dose planning and help identify patients who could benefit from combined radiotherapy, chemotherapy, targeted therapy, or immunotherapy.

The current priority in breast cancer radiotherapy is to reduce cardiac doses without compromising target tissue coverage. The voluntary breath-hold technique described here is a simple, inexpensive solution to this problem and capable of being instituted widely without the need for specialized equipment.

Brain tumor patients routinely undergo cranial radiotherapy, and while beneficial, this treatment often results in debilitating cognitive dysfunction. This serious unresolved problem has at present, no clinical recourse, and has driven our efforts to devise stem cell based therapies for the recovery of radiation-induced cognitive decrements.

We describe a rat model of post traumatic stress disorder (PTSD) that reveals the persistent alterations in neuroendocrine function and the delayed long-term, exaggerated fear response, characteristic of PTSD patients. The animal model and methods described here are useful for correlating biomarkers in brain nuclei, which are mechanistic but cannot be measured in patients, with biomarkers in peripheral white blood cells, which can.
