Tissue Punch Technique

The tissue punch technique is a sampling method that removes a defined, cylindrical core of tissue for microscopic, histological, or molecular analysis. A hollow, sharp-edged punch is positioned over the target region and pressed or rotated through the specimen, producing a standardized tissue core while preserving its spatial context. In cancer research, tissue punches can isolate tumor and adjacent normal regions from surgical specimens, tissue blocks, or experimental models for DNA, RNA, protein, and biomarker studies. Consistent sampling supports tissue microarray construction, comparison among tumors, and more reproducible evaluation of cancer biology, treatment response, and disease progression.

The fibroblast explant culture protocol from human skin punch biopsies is a technically robust and simple way to derive skin cells within 4-8 weeks for banking of about 15-20 million cells at a low passage number.

A Technique for the Prolonged Incubation of Neuronal Tissue of the Retina

2025

This video demonstrates a method for prolonged incubation of neuronal tissue of the retina. Eyecups containing retina are obtained from a mouse eye; the inner limiting membrane (ILM) is enzymatically digested to expose the neuronal cells of the retina, and either the treated eyecup or the isolated retina is transferred to a specialized incubator for prolonged incubation at a low temperature.
