Alpha Position

The alpha position in chemistry denotes the site immediately adjacent to a reference functional group or reactive center, most commonly the carbon next to a carbonyl group in organic molecules. In carbonyl compounds, the alpha carbon may bear acidic alpha hydrogens; removal of one by a base forms a resonance-stabilized enolate, which can act as a nucleophile in reactions such as alkylation and aldol condensation. Understanding alpha-position reactivity helps predict reaction pathways, select suitable conditions, and design synthetic strategies for building carbon-carbon bonds. The term also supports structural analysis and nomenclature in broader organic and biochemical contexts.

This video demonstrates an immunofluorescence assay to detect neurodegenerative disease-specific alpha-synuclein aggregates on skin biopsy sections. Free-floating immunostaining is employed to fluorescently label the carboxyl-terminal hydrolase and the alpha-synuclein aggregates in the dermal neurons. Under a confocal microscope, cells with fluorescence from both antibodies indicate the presence of alpha-synuclein aggregates in the dermal neurons.
