Current Efficiency Optimization

Current efficiency optimization is the improvement of an electrochemical process so that the applied electrical current produces the desired chemical transformation with minimal waste. It relies on controlling electrode potential, current density, electrolyte composition, temperature, and mass transport to favor the target reaction while suppressing competing pathways, such as solvent decomposition or side-product formation. In chemistry, optimized current efficiency can reduce energy and material consumption, improve product selectivity, and increase process yield in electrosynthesis, electroplating, and electrochemical manufacturing. Measuring charge passed against product formed also provides a practical basis for comparing reactor designs and operating conditions.

We present our optimized high-throughput nucleofection protocol as an efficient way of transfecting primary human monocyte-derived dendritic cells with either plasmid DNA or siRNA without causing cell maturation. We further provide evidence for successful siRNA silencing of targeted gene RIG-I at both the mRNA and protein levels.
