Intradermal Delivery

Intradermal delivery is a drug administration method that deposits a therapeutic, vaccine, or diagnostic agent within the dermis, the skin layer beneath the epidermis. A shallow needle or specialized device places a small volume at a controlled depth, where the agent interacts with resident immune cells and the dense network of dermal microvessels and lymphatic vessels. This localized route can support immune stimulation, regional treatment, and diagnostic testing while potentially reducing systemic exposure or dose requirements. In clinical research, intradermal delivery is evaluated for vaccination, allergy testing, local anesthesia, and emerging approaches that use the skin as an accessible interface for targeted therapy.

Myeloid Cell Isolation from Mouse Skin and Draining Lymph Node Following Intradermal Immunization with Live Attenuated Plasmodium Sporozoites

We describe here a protocol for isolating myeloid cells from mouse skin and draining lymph node following intradermal injection of Plasmodium sporozoites. Flow cytometry of collected cells provides a reliable assay to characterize the skin and draining lymph node inflammatory response to the parasite.

Skin tattooing is a potent and safe way to delivery DNA vaccine intradermally. Here, a DNA plasmid encoding EGFP is delivered by tattooing to the skin of a laboratory mouse, and the expression of EGFP in the skin cells is then inspected by confocal microscopy.
