Cd34-tta/teto-h2bgfp Mice

Cd34-tta/teto-H2BGFP mice are genetically engineered reporter animals used to identify and track Cd34-expressing cells and their descendants during development and tissue maintenance. In this Tet-Off system, the Cd34 promoter drives tetracycline transactivator expression, which binds tetracycline response elements to activate a histone H2B-GFP reporter; doxycycline suppresses labeling, while subsequent cell divisions dilute the nuclear GFP signal. Researchers can therefore distinguish relatively quiescent or slowly cycling cells from rapidly proliferating populations and follow changes in cell behavior over time. This model supports studies of stem and progenitor cell dynamics, lineage persistence, tissue regeneration, and developmental processes in vivo.

This study has established a stable and efficient method for the isolation, culture, and functional determination of vascular wall-resident CD34+ stem cells (CD34+ VW-SCs). This easy-to-follow and time-effective isolation method can be utilized by other investigators to study the potential mechanisms involved in cardiovascular diseases.

Isolating Cord Blood Hematopoietic Stem Cells to Generate Human Immune System Mice

This video demonstrates a technique for generating human immune system (HIS) mice. CD34+ hematopoietic stem cells (HSCs) are isolated from human cord blood buffy coat cells using antibody-coated magnetic beads. The CD34+ HSCs are expanded in a growth medium enriched with cytokines and cryopreserved. Finally, the thawed HSCs are injected into an immunodeficient mouse pup to establish the HIS mouse model.

This procedure is useful for in-depth analysis of cellular and molecular mechanisms controlling megakaryopoiesis.

Here, we present a protocol for immunophenotypic characterization and cytokine induced differentiation of cord blood derived CD34+ hematopoietic stem and progenitor cells to the four myeloid lineages. The applications of this protocol include investigations on the effect of myeloid disease mutations or small molecules on myeloid differentiation of the CD34+ cells.
