Colony Manual Dissection

Colony manual dissection is a technique for mechanically separating selected regions of a cell colony under microscopic guidance, allowing researchers to preserve and transfer defined groups of cells. In developmental biology, a fine needle or similar tool is used to cut an adherent colony into small fragments, which are then collected and replated under conditions that support continued growth. This approach enables controlled expansion of stem or progenitor cell populations while limiting unwanted differentiation and contamination from neighboring regions. It is useful for maintaining cell lines, isolating colonies with distinct morphologies, and studying how cellular organization and developmental potential change during culture.

In this video, we perform a clonogenic assay of keratinocytes harvested from adult mice to determine the ability of individual cells to proliferate and form colonies. This keratinocyte harvesting technique is useful for studying other downstream aspects of cutaneous chemical carcinogenesis and tumor promotion.
