Floating Culture Conditions

Floating culture conditions are laboratory settings in which cells, tissues, or developing structures remain suspended in culture medium rather than attaching to a solid substrate. Researchers typically use nonadherent surfaces, specialized vessels, or controlled agitation to prevent attachment while supplying nutrients, gases, and signaling molecules needed for growth. In developmental biology, this arrangement can support three-dimensional cell aggregation and tissue organization, allowing cells to interact across multiple surfaces during differentiation. Floating cultures are used to study morphogenesis, lineage specification, embryoid body formation, and organoid development, while helping researchers examine how cellular interactions and environmental conditions influence developmental outcomes.

A protocol to prepare free-floating slice cultures from adult human brain is presented. The protocol is a variation of the widely used slice culture method using membrane inserts. It is simple, cost-effective, and recommended for running short-term assays aimed to unravel mechanisms of neurodegeneration behind age-associated brain diseases.
