Nucleoside Labeling

Nucleoside labeling is a method for tracking nucleic acid synthesis by supplying cells with detectable nucleoside analogs that become incorporated into newly formed DNA or RNA. During DNA replication, proliferating cells take up labels such as bromodeoxyuridine or ethynyl deoxyuridine, which can be detected through antibody-based staining or chemical click reactions. In developmental biology, these signals reveal when and where cells divide, helping researchers map growth patterns, analyze progenitor-cell behavior, and follow cell-cycle dynamics across tissues and developmental stages. The approach links molecular synthesis to cell proliferation and supports studies of tissue formation, regeneration, and developmental disease.

Here, we present protocols for the synthesis of disaccharide nucleosides by the regioselective O-glycosylation of ribonucleosides via a temporary protection of their 2',3'-diol moieties utilizing a cyclic boronic ester. This method applies to several unprotected nucleosides such as adenosine, guanosine, cytidine, uridine, 5-methyluridine, and 5-fluorouridine to give corresponding disaccharide nucleosides.
