Porcine Subcutaneous Adipose Tissue

Porcine subcutaneous adipose tissue is the fat depot located beneath the skin of pigs, where adipocytes store energy and connective and vascular cells support tissue function. During development, mesenchymal progenitor cells proliferate and commit to the adipocyte lineage, then accumulate lipid as insulin, glucocorticoids, nutrition, and local signals regulate adipogenesis and tissue expansion. Studying this tissue helps developmental biologists connect cellular differentiation with whole-animal growth, body composition, and metabolic regulation. Because pigs share important anatomical and physiological features with humans, porcine subcutaneous adipose tissue provides a valuable model for investigating adipose development, obesity-related biology, and factors that influence meat quality.

Isolation and Differentiation of Adipose-Derived Stem Cells from Porcine Subcutaneous Adipose Tissues

This protocol describes the isolation of pig adipose-derived stem cells (pADSC) from subcutaneous adipose tissues with examination of multipotency. The multipotent pADSC are used to delineate processes of adipocyte differentiation and study transdifferentiation into multiple cell lineages of mesodermal mesenchyme or further lineages of ectoderm and endoderm for regenerative studies.

A Technique for Subcutaneous Abdominal Adipose Tissue Biopsy via a Non-diathermy Method

We standardized an abdominal adipose tissue biopsy using a non-diathermy method performed under local anesthesia. Three cases of excessive post-operation bleeding out of 115 operations (2.61%) occurred.We conclude that an abdominal adipose tissue surgical biopsy using a non-diathermy method can be safely applied to healthy men.

The manuscript and associated video demonstrate a percutaneous biopsy technique to obtain samples of subcutaneous adipose tissue from areas surrounding the umbilicus. This method is a low-risk and efficient way to investigate a range of parameters (e.g., gene or protein expression, enzyme activity, lipid content) within adipose tissue.
