Cryogenic Milling

Cryogenic milling is a size-reduction technique that uses extremely low temperatures to process materials that are difficult to grind under conventional conditions. The material is cooled, commonly with liquid nitrogen, until it becomes brittle, then fractured by impact, attrition, or cutting in a mill; cooling also limits heat generation, melting, oxidation, and degradation. In engineering, this method produces fine, uniform powders from polymers, elastomers, pharmaceuticals, foods, and temperature-sensitive composites. Cryogenic milling can improve particle-size control, preserve material properties, and support powder processing, recycling, additive manufacturing, and the production of specialized research materials.

Workflow Using a Cryogenic Coincident Fluorescence, Electron, and Ion Beam Microscope for Targeted Milling of Cells

This workflow enables lamella production targeting fluorescently labeled biological structures that are small (<1 μm in axial extent) and rare (1 copy per cell) using a cryogenic tri-coincident imaging platform. This platform integrates fluorescence microscopy, focused ion beam milling, and scanning electron microscopy at a single focal position and enables simultaneous fluorescence microscopy while milling.
