Surface Potential Microscopy

Surface Potential Microscopy is a scanning probe technique that maps local electrical potential across a material surface, revealing variations that influence device performance and interfacial behavior. In a common implementation, a conductive atomic force microscope tip scans the sample while an applied voltage and controlled tip vibration generate electrostatic interactions; feedback adjusts the voltage to null the electrostatic force, measuring the contact potential difference between tip and surface. In engineering research, these maps help characterize semiconductors, photovoltaic materials, thin films, corrosion, and electronic interfaces, linking nanoscale charge distribution to defects, work function, contamination, and electrical performance.

This protocol shows a plant sample preparation method for light-sheet microscopy. The setup is characterized by mounting the plant vertically on the surface of a gel and letting it grow in controlled bright conditions. This allows long-term observation of plant organ development in standardized conditions.

Mitochondria can utilize the electrochemical potential across their inner membrane (ΔΨm) to sequester calcium (Ca2+), allowing them to shape cytosolic Ca2+ signaling within the cell. We describe a method for simultaneously measuring mitochondria Ca2+ uptake and ΔΨm in live cells using fluorescent dyes and confocal microscopy.

Microscopy-Based Evaluation of Bacterial Attachment to Plant Root Surfaces

Source: Matthysse, A. G. Adherence of Bacteria to Plant Surfaces Measured in the Laboratory. J. Vis. Exp. (2018).This video demonstrates the evaluation of bacterial attachment to plant roots using phase-contrast microscopy. Root samples from seedlings incubated with bacteria are first mounted to check for free bacteria. The samples are then washed to remove loosely attached bacteria and remounted using a suitable coverslip to prevent damage. Finally, the number of bacteria attached to the root...

Source: Yong P. Chen, PhD, Department of Physics & Astronomy, College of Science, Purdue University, West Lafayette, IN Electric potential, also known as "voltage", measures the electric potential energy per unit charge. Electric field is a scalar quantity and is fundamental to many electrical effects. Like potential energy, what is physically meaningful is the difference in the electric potential. For example, the spatial variation in the electric potential is related to the electric...
