Enrichment Expansion Technique

The Enrichment Expansion Technique is a laboratory strategy for increasing the proportion and quantity of a desired biological population through selective enrichment followed by controlled expansion. During enrichment, culture conditions or selection steps favor target cells, microorganisms, or other engineered entities over unwanted populations; expansion then propagates the enriched material to obtain sufficient biomass or cell numbers for analysis and use. In engineering research, this approach supports the development of cell-based systems, microbial bioprocesses, and other biological platforms that require consistent, high-yield populations. By improving population purity and scale, the technique helps connect discovery-stage selection with downstream characterization, manufacturing, and performance testing.

This video describes a protocol for isolating rare-antigen-specific CD8+ T cells using magnetic nanoparticle-based artificial antigen-presenting cells (aAPCs) functionalized with antigenic peptide-loaded MHC-Igs and anti-CD28 antibodies. Interaction of the aAPCs with CD8+ T cells specifically expressing the antigenic peptide-targeting T cell receptors, followed by their expansion in vitro, results in the enrichment and expansion of this CD8+ T cell population.

Expansion and Enrichment of Gamma-Delta (γδ) T Cells from Apheresed Human Product

Presented is a protocol for the expansion of Gamma-Delta (γδ) T cell drug product. Lymphocytes are isolated by elutriation and γδ-enriched with zoledronic acid and interleukin-2. Alpha-beta T cells are depleted using a clinical-grade magnetic separation device. The γδ cells are co-cultured with K562-derived, artificial antigen-presenting cells and expanded.

We present an enrichment protocol for the isolation of bacteriophages infecting bacteria in the Arthrobacter genus. This enrichment protocol produces fast and reproducible results for the isolation and amplification of Arthrobacter phages from soil isolates.
