Fault Localization

Fault localization is the systematic process of identifying where a failure or abnormal behavior originates within an engineered system, a critical step in restoring safe, reliable operation. It works by comparing observed symptoms, measurements, or test results with expected system behavior, then tracing discrepancies through components, connections, subsystems, or software modules to isolate the likely fault location. Engineers apply fault localization in electrical networks, mechanical equipment, control systems, and software debugging to reduce downtime, guide repairs, and validate designs. Methods such as signal analysis, diagnostic testing, redundancy checks, and model-based reasoning also support predictive maintenance and improve system safety.

We describe a method to localize fluorescently tagged proteins in electron micrographs. Fluorescence is first localized using photo-activated localization microscopy on ultrathin sections. These images are then aligned to electron micrographs of the same section.
