Gel Pores

Gel pores are nanoscale or microscale voids within a gel network that contain liquid or gas and influence the material’s structure and performance. They form when polymer chains, particles, or reaction products assemble into a three-dimensional matrix, while solvent retention, swelling, drying, and cross-linking determine pore size and connectivity. In engineering, gel pores affect permeability, diffusion, mechanical strength, water transport, and dimensional stability in materials such as hydrogels, cementitious systems, coatings, and porous composites. Characterizing their distribution helps researchers control material processing and optimize systems for filtration, tissue engineering, drug delivery, sensing, and structural applications.

Gel Purification

2023

Gel purification is used to recover DNA fragments after electrophoretic separation. DNA recovery from an agarose gel includes three basic steps: binding, washing and eluting from a silica column. DNA is believed to bind to silica in the presence of high salt via a salt bridge. Following binding, DNA is washed of impurities and eluted under low salt conditions disrupting this interaction. This video goes through a step-by-step, generalized procedure for cutting out a band from the gel, gel...

Pore Size Distribution

2024

In concrete, the pore size distribution significantly influences the material's properties. Capillary pores, markedly larger than gel pores, form a vast network within partially hydrated cement paste, reducing the concrete's strength and increasing its permeability. This heightened permeability leads to a greater risk of damage from environmental factors like freeze-thaw cycles and chemical attacks, with the extent of vulnerability also being tied to the water-to-cement ratio. Adequate...

2014

A procedure for creating and imaging capillary bridges in slit-pore geometry is presented. The creation of capillary bridges relies on the formation of pillars to provide a directional physical and chemical heterogeneity to pin the fluid. Capillary bridges are formed and manipulated using microstages and visualized using a CCD camera.

DNA Gel Electrophoresis

2023

DNA gel electrophoresis is a technique used for the detection and separation of DNA molecules. An electric field is applied to a gel matrix comprised of agarose, and within the gel, charge particles will migrate and separate based on size. The negatively charged phosphates of the DNA backbone cause DNA fragments to move toward the anode - a positively charged electrode. The video explains the mechanism by which DNA fragments are resolved on an agarose gel, and it provides a step-by-step...
