Pupae Handling Technique

Pupae handling technique is the controlled collection, transfer, and maintenance of insect pupae, an important practice for preserving specimens and obtaining reliable developmental observations. Because pupae undergo metamorphosis while remaining largely immobile, careful manipulation, suitable humidity and temperature, clean containers, and protection from compression or desiccation help support normal adult emergence. In environmental research, consistent pupal handling improves insect rearing, life-cycle studies, population monitoring, and investigations of how environmental conditions affect development. Standardized procedures also reduce mortality and variation, strengthening comparisons among treatments, habitats, or species and supporting applications in ecology, conservation, and environmentally focused biological research.

The video demonstrates the use of photoconversion to study inflammatory cell dynamics in Drosophila pupae. Green hemocytes are drawn to the wounded epithelium, followed by distant migration. Photoconversion shifts certain hemocytes from green to red, facilitating their movement tracking.

RNA interference (RNAi) is an extremely valuable tool for uncovering gene function. However, the ability to target genes using RNAi during pre-adult stages is limited in the major human malaria vector Anopheles gambiae. We describe an RNAi protocol to reduce gene function via direct injection during pupal development.
