Parental Rnai

Parental RNAi (RNA interference) is a genetic method in which double-stranded RNA introduced into a reproducing organism reduces expression of a matching gene in its progeny, making it useful for studying gene function across generations. After uptake, cellular enzymes process the double-stranded RNA into small interfering RNAs that guide an Argonaute-containing silencing complex to complementary messenger RNA, promoting its degradation or blocking translation. In model organisms such as nematodes, parental RNAi can produce gene knockdown in embryos and larvae without directly treating each offspring. The approach supports functional genomics, developmental studies, and analysis of inherited gene-silencing effects.

RNA interference (RNAi) is a widely used technique in which double stranded RNA is exogenously introduced into an organism, causing knockdown of a target gene. In the nematode, C. elegans, RNAi is particularly easy and effective because it can be delivered simply by feeding the worms bacteria that express double stranded RNA (dsRNA) that is complementary to a gene of interest. First, this video will introduce the concept of RNA interference and explain how it causes targeted gene knockdown.

Bacterial Delivery of RNAi Effectors: Transkingdom RNAi

For development of RNA interference (RNAi)-based therapies, a novel strategy was developed, transkingdom RNAi (tkRNAi). This technology uses non-pathogenic bacteria to produce and deliver therapeutic short hairpin RNA (shRNA) into target cells. Here, tkRNAi was successfully applied for reversal of classical ABCB1-mediated multidrug resistance (MDR) of cancer cells.
