Patient Tissue Samples

Patient tissue samples are biological specimens collected from individuals for laboratory analysis, providing direct molecular and cellular evidence of health, disease, and inherited variation. In genetics research, tissue is obtained through biopsy or surgery, preserved to maintain its integrity, and processed to extract DNA or RNA for methods such as sequencing, genotyping, or gene-expression analysis. These samples help researchers identify inherited variants, somatic mutations, and molecular patterns associated with cancer and other conditions. When linked with clinical information and handled with appropriate consent and privacy protections, patient tissue samples support diagnosis, disease classification, treatment research, and the development of more precise genetic medicine.

Circulating tumor cells are isolated from the blood of cancer patients without inflicting cellular damage. Isolation of tumor cells is accomplished using a bimolecular surface of E-selectin in addition to antibodies against epithelial markers. A nanotube coating specifically promotes cancer cell adhesion resulting in high capture purities.

Quantitation of DNA double-strand breaks on the basis of γH2AX foci has become an invaluable tool, particularly in radiation biology, for the evaluation of tissue radiosensitivity and effects of radiation modifying compounds. Here we demonstrate the use of an immunofluorescence assay for quantitation of γH2AX foci in tissue samples.

A Hydrophobic Tissue Clearing Method for a Rat Brain Tissue Sample

Source: Kirchner, K.N., et al., A Hydrophobic Tissue Clearing Method for Rat Brain Tissue. J. Vis. Exp. (2020).This video demonstrates the method of hydrophobic tissue clearing of a rat brain tissue sample. The sample is sequentially dehydrated in methanol, treated with a dichloromethane and methanol mixture to dissolve lipids and pigments, and transferred to dibenzyl ether for complete tissue clearing, enabling detailed imaging without thin sectioning.
