Bal Fluid Analysis

BAL fluid analysis is the laboratory examination of bronchoalveolar lavage fluid collected from the lower respiratory tract to assess inflammation, infection, and lung injury. During bronchoscopy, sterile saline is introduced into a selected airway and gently aspirated; the recovered fluid is then evaluated for immune cells, microorganisms, proteins, and other cellular or molecular markers. In immunology and infection research, these measurements help characterize host responses, distinguish infectious from noninfectious pulmonary disease, and monitor disease progression or treatment. The approach also supports studies of airway immunity by linking local cellular and microbial findings with respiratory symptoms and clinical outcomes.

Bronchoalveolar Lavage (BAL) for Research; Obtaining Adequate Sample Yield

We describe a research technique for fiberoptic bronchoscopy and bronchoalveolar lavage using low pressure suction. The technique is used to harvest immune cells from the lung bronchoalveolar surfaces. Local anesthetic and mild conscious sedation (midazolam) is used. Subjects tolerate the procedure well and experience minimal side effects.

Sample Preparation for Endopeptidomic Analysis in Human Cerebrospinal Fluid

A method for mass spectrometric analysis of endogenous peptides in human cerebrospinal fluid (CSF) is presented. By employing molecular weight cut-off filtration, chromatographic pre-fractionation, mass spectrometric analysis and a subsequent combination of peptide identification strategies, it was possible to expand the known CSF peptidome nearly ten-fold compared to previous studies.

Repeated measurements of rodent respiratory physiology and sampling of airway inflammatory cells are desirable, but generally not feasible. Here we describe a repeatable method for orally intubating mice that permits repeated measurements of airway hyperreactivity and sampling of airway inflammatory cells.

Synovial fluid analysis under transmitted, polarized, and compensated light microscopy is used to evaluate the inflammatory or non-inflammatory nature of a sample through simple steps. It is particularly useful in osteoarthritis to detect calcium crystals and identify a more severe subset of osteoarthritis.
