Blocking Step

The blocking step is a preparatory procedure in immunological assays that reduces nonspecific interactions between assay surfaces and detection reagents, improving the reliability of measured immune signals. It works by coating unoccupied binding sites, commonly on a plate or membrane, with an inert protein or other blocking reagent before the antigen, antibody, or sample is added. In ELISA, Western blotting, and related infection research methods, effective blocking lowers background signal and helps distinguish specific antigen–antibody binding from incidental adsorption. Selecting a compatible blocker and optimizing its concentration and incubation conditions can improve assay sensitivity, reproducibility, and interpretation.
