Cd4 Cd8 Double Positive

CD4/CD8 double-positive (DP) thymocytes are immature T cells that express both CD4 and CD8 coreceptors during a critical stage of thymic development. After T-cell receptor rearrangement, their receptors are tested against self-peptide–MHC complexes: productive recognition promotes survival and lineage commitment to CD4 or CD8 single-positive cells, whereas weak or excessively strong interactions lead to death by neglect or negative selection. Studying DP cells with flow cytometry, tissue analysis, and genetic models helps define T-cell selection, central tolerance, and immune development. These cells provide a framework for understanding thymic dysfunction, immunodeficiency, autoimmunity, and altered T-cell responses during infection.

Generating Single Positive CD8 T Cells from Induced Pluripotent Stem Cells

This video demonstrates an assay to generate tumor antigen-specific induced pluripotent stem cell (iPSC)-derived CD8αβ+ T Cells. Tumor antigen-specific iPSCs are grown on a stromal cell monolayer and differentiated into hematopoietic progenitor cells (HPCs). The HPCs are differentiated into CD4+ CD8+ double-positive T cells using a differentiation medium. These cells are then stimulated to generate single-positive T cells expressing CD8.

An Assay for Cell Cycle Analysis of Antigen-Specific CD8+ T Cells

This video demonstrates an assay to analyze antigen-specific CD8+ T cells in different cell cycle stages. Fluorescently-labeled viable cells are selected using flow cytometry, and the cells in the different cell cycle stages are identified by their DNA content using antibodies against cell proliferation-associated nuclear protein and a nucleic acid stain.

This video demonstrates a protocol for the magnetic labeling-based isolation of a low-concentration population of CD4+Vδ1+γδ T cells from human peripheral blood mononuclear cells (PBMCs). Using a combination of FITC-conjugated anti-Vδ1 antibodies, magnetic anti-FITC microbeads, and anti-CD4 antibody-coated superparamagnetic beads, the CD4+Vδ1+ T-cells can be successfully purified from the PBMC population.

This video describes a protocol for isolating rare-antigen-specific CD8+ T cells using magnetic nanoparticle-based artificial antigen-presenting cells (aAPCs) functionalized with antigenic peptide-loaded MHC-Igs and anti-CD28 antibodies. Interaction of the aAPCs with CD8+ T cells specifically expressing the antigenic peptide-targeting T cell receptors, followed by their expansion in vitro, results in the enrichment and expansion of this CD8+ T cell population.
