Expression Constructs

Expression constructs are engineered DNA molecules that direct production of a selected gene product in a host cell, making them essential tools for studying immune responses and infectious diseases. They typically combine a promoter, coding sequence, and regulatory elements that control transcription, followed by translation of the resulting messenger RNA into a protein. In immunology and infection research, these constructs can produce recombinant antigens, antibodies, cytokines, or pathogen proteins for functional assays, diagnostics, vaccine studies, and mechanistic investigations. Their design influences expression level, cellular localization, protein stability, and experimental reproducibility, supporting both basic research and development of immune-based interventions.

Lentiviral expression vectors are the most effective vehicles for stably expressing different effector molecules or reporter constructs in dividing and non-dividing mammalian cells and whole organisms. Here we provide a protocol on how to package lentivector expression constructs in pseudoviral particles and to transduce target cells using the pseudoviral particles.

We describe here a protocol for the generation of iCMs using retrovirus-mediated delivery of Gata4, Tbx5 and Mef2c in a polycistronic construct. This protocol yields a relatively homogeneous population of reprogrammed cells with improved efficiency and quality and is valuable for future studies of iCM reprogramming.
