Extracellular Myeloperoxidase

Extracellular myeloperoxidase is the heme enzyme released by activated neutrophils and monocytes into the extracellular environment, where it contributes to antimicrobial defense and inflammatory injury. During degranulation or neutrophil extracellular trap formation, myeloperoxidase uses hydrogen peroxide and chloride ions to generate hypochlorous acid, a potent oxidant that damages microbial proteins, membranes, and other cellular components. This activity helps control bacterial and fungal infections, but excessive or poorly localized oxidation can injure host tissues and amplify inflammation. Measuring extracellular myeloperoxidase therefore supports research on innate immunity, infection, inflammatory disease, and the balance between pathogen killing and collateral tissue damage.

We present a protocol for a modified sandwich enzyme-linked immunosorbent assay technique to quantitatively measure two components of neutrophil extracellular trap remnants, myeloperoxidase conjugated-DNA and neutrophil elastase conjugated-DNA complexes, derived from activated neutrophils.

Induction and Visualization of Neutrophil Extracellular Traps in Pre-Labeled Neutrophils

2025

This video demonstrates the induction and visualization of neutrophil extracellular traps or NETs in pre-labeled neutrophils using patient serum containing anti-neutrophil cytoplasmic antibodies. Furthermore, using an impermeable DNA dye and fluorescence microscopy enables the visualization of NETs.
