Graft Bed Preparation

Graft bed preparation is the process of conditioning a recipient wound so transplanted tissue can attach, survive, and heal. It typically involves removing necrotic tissue and contaminants, controlling infection and bleeding, and establishing a clean, vascularized surface that supports nutrient and oxygen exchange; these steps also limit microbial persistence and excessive inflammation. In immunology and infection research, graft bed preparation helps clarify how host immune responses, local cytokines, and tissue vascularity influence graft take. Clinically, careful preparation improves healing outcomes and reduces graft loss in burns, traumatic wounds, and chronic ulcers.

This video demonstrates a method for grafting human skin onto an immunodeficient mouse with a suppressed neutrophil response. The method involves applying human split-thickness skin to the mouse's graft bed, sealing it, and covering it for recovery. Eventually, the human skin adheres to the mouse's skin, establishing vascularization and circulation, resulting in a successful skin xenograft.
