Ipec-j2 Cells

IPEC-J2 cells are non-transformed porcine jejunal epithelial cells used as an in vitro model of the intestinal lining, particularly in immunology and infection research. When cultured under appropriate conditions, they form polarized monolayers with apical and basolateral surfaces, tight junctions, and measurable transepithelial electrical resistance that reproduce key features of the epithelial barrier. Researchers use IPEC-J2 cells to investigate pathogen attachment and invasion, epithelial permeability, microbial translocation, and host responses to infection or inflammatory stimuli. Their controlled, reproducible format supports studies of intestinal barrier function, epithelial-immune interactions, and potential strategies for preventing or treating enteric disease.

Zika Virus (ZIKV), an emerging pathogen, is linked to fetal developmental abnormalities and microcephaly. The establishment of an effective infectious cell culture system is crucial for studies of ZIKV replication as well as vaccine and drug development. This protocol details the derivation of transplantable hematopoietic stem cells from mouse embryonic stem cells (ESC) and their subsequent injection into lethally irradiated recipient mice. Briefly, ESC are differentiated as embryoid bodies, which are then infected with retroviral HoxB4 and co-cultured with OP9 stromal cells and hematopoietic cytokines.
