Mdck Cell Overlay

An MDCK cell overlay is a semisolid culture layer placed over Madin-Darby Canine Kidney (MDCK) cell monolayers to localize viral spread and make infection patterns measurable. In plaque assays, susceptible MDCK cells are infected with virus, then covered with agarose- or methylcellulose-containing medium that limits diffusion of progeny virions to neighboring cells, producing discrete zones of cell damage called plaques. After incubation and staining, researchers count plaques to estimate infectious virus titer and compare replication, neutralization, or antiviral activity. This approach supports influenza and other respiratory virus studies by linking visible infection outcomes to quantitative virology.

Plaque assays are the gold standard for viral quantification, utilizing entrapping overlays on host cellular monolayers to determine viral titers. While various semisolid overlays have traditionally been used, here we demonstrate plaque techniques comparing semisolid overlays to a novel liquid microcrystalline cellulose among several families of viruses.

Testing protein-protein interaction is indispensable for dissection of protein functionality. Here, we introduce an in vitro protein-protein binding assay to probe a membrane-immobilized protein with a soluble protein. This assay provides a reliable method to test interaction between an insoluble protein and a protein in solution.

(2017)This video demonstrates the procedure for detecting bacteriocin-mediated killing by spotting supernatant from an antibiotic-induced donor strain onto a soft agar overlay containing a test strain. Clear zones formed after incubation indicate bacterial inhibition.
