Nascent Rna

Nascent RNA is newly synthesized RNA produced during active transcription, providing a direct view of gene expression as it occurs. RNA polymerases generate nascent transcripts from DNA templates, and these short, emerging molecules can be captured through metabolic labeling, run-on assays, or sequencing approaches that distinguish new RNA from preexisting transcripts. In immunology and infection research, nascent RNA analysis reveals how immune cells rapidly activate or silence genes after pathogen recognition, cytokine stimulation, or antiviral signaling. These measurements help separate transcriptional regulation from changes in RNA stability, improving understanding of host responses, pathogen-driven gene control, and potential therapeutic targets.

Trophoblast giant cells (TGCs) play a key role in the placenta to ensure a healthy pregnancy. We present a protocol for assessing the transcriptional status of genes in TGCs by nascent fluorescent in situ hybridization on cryostat sections of post-implantation embryos or short-term cultures of embryonic day 7 ectoplacental cones.

Isolation of Ribosome Bound Nascent Polypeptides in vitro to Identify Translational Pause Sites Along mRNA

A technique to identify translational pause sites on mRNA is described. This procedure is based on isolation of nascent polypeptides accumulating on ribosomes during in vitro translation of a target mRNA, followed by the size analysis of the nascent chains using a denaturing gel electrophoresis.
