Natural Killer Cell Cytotoxicity

Natural killer (NK) cell cytotoxicity is the ability of NK cells to identify and destroy stressed, infected, or malignant cells without prior antigen-specific sensitization, making it an important component of innate immunity. NK cells integrate signals from activating and inhibitory receptors, including reduced major histocompatibility complex class I expression, then form an immunological synapse and release perforin and granzymes that trigger target-cell apoptosis; they can also engage death receptors. In immunology and infection research, measuring NK cytotoxicity helps assess antiviral defense, immune dysfunction, and responses to cancer therapies. Understanding this process supports studies of host-pathogen interactions and immune-based treatments.

This video demonstrates an in vitro technique to measure the cytotoxic ability of cytokine-induced killer (CIK) cells against cancer cells. Upon co-culturing target cancer cells with CIK cells, the dead cancer cells are identified using a fluorescent viability dye and are quantified using flow cytometry, estimating the cytotoxic effect of CIK cells.
