Macrophages Microglia

Macrophages and microglia are tissue-resident phagocytic cells that provide rapid innate immune defense, with microglia serving as the macrophage-like immune cells of the central nervous system. Through pattern-recognition receptors, they detect pathogen-associated or damage-associated signals, engulf microbes and cellular debris, and release cytokines and chemokines that shape inflammation; they can also present antigens to adaptive immune cells. Studying these cells in immunology and infection clarifies how tissues contain pathogens, repair injury, or develop harmful inflammation, including neuroinflammatory responses. Their activation is therefore important for understanding infectious disease, chronic inflammation, and therapeutic design.

Here we describe two methods to study glioblastoma cell interaction with tumor associated macrophages and microglia where the effect on glioblastoma invasion is assessed.

We describe immunofluorescence-based protocols followed by three-dimensional confocal analysis to investigate the pattern of co-expression of microglia/macrophage phenotype markers in a mouse model of focal ischemia.
