Peptide Elution

Peptide elution is a method for releasing peptides bound to major histocompatibility complex (MHC) molecules so their identities and immune significance can be determined. Typically, MHC–peptide complexes are isolated from cells or tissues, exposed to acidic conditions that disrupt noncovalent interactions, and separated from the released peptides for analysis by liquid chromatography and mass spectrometry. In immunology and infection research, this approach reveals pathogen-derived and host-derived epitopes displayed on infected or antigen-presenting cells, helping researchers define T-cell targets, assess antigen processing, and guide vaccine or immunotherapy development.

A miniature column containing resin beads bonded to long hydrocarbon chains facilitates hydrophobic interaction-based purification. Upon loading a pre-digested peptide sample into the column, the peptide fragments bind to the resin beads, allowing impurities to pass through. An elution buffer that disrupts...

High-Performance Liquid Chromatography: Elution Process

2024

In High-Performance Liquid Chromatography (HPLC), the elution process is critical to the separation of analytes and the quality of chromatographic results. Elution describes how compounds move through the column and separate based on their interactions with the mobile and stationary phases. This process determines the resolution, peak shape, and retention times in the chromatogram, which are essential for identifying and quantifying components in complex mixtures. Understanding the elution...
